Cloning and Sequence Analysis of the Gene rps7 Coding for Chloroplast Ribosomal Protein CS7 from Brassica napus

A 1. 0 kb DNA fragment containing the rps7 gene for ribosomal protein CS7 was cloned from chloro-plast genome of rape (Brassica napus) cv. H165. The sequence analysis results indicated that the coding region of rps7 gene contained 468 nucleotides coding a protein of 155 amino acids, with a calculated molecular weight of 20 109 D. Its DNA and deduced amino acid shared sequence homologies of 97% and 97%, respectively, with those of tabacco, while 90% and 84%, respective-ly, with those of rice. None of the introns was present in this gene. Although typical Shine2Dalgarno (SD) se-quence was not found within the 28 bp upstreamfrom the start codon, a conserved TGAT box (at-25 to-22), which was identified as an essential cis2element (RBS2) for mRNA translation of psbA in tobacco, was detected. Compared with the homogeneous sequences from tobacco and rice, insertions and deletions frequently occur at its 3'2 non2coding region. In addition, the restriction en-donuclease map of the cloned DNA was also worked out. The sequence data reported here have been deposited in GenBank.
