论文摘要:Protoplast isolation is relevant, for many different,applications, and has been principally used in procedures involving genetic manipulation. In this study, the age of mycelium, osmotic stabilizers, enzyme, incubation temperature and incubation time were evaluated in terms of their effects on protoplast yield. The yong mycelia (3d) of Tulasnella calospora were digested for 6h at 30℃ in a mixture of 1.2 mol・L MgSO4 +10mmol・L-1 K2HPO4 as the osmotic stabilizer, with a 1.0% lysing enzyme and 1.5% driselase: more than 106 protoplasts mL-1 were obtained. When collected by density gradient centrifugation, the concentration of protoplasts can reach 107-108 protoplasts mL-1, an amount suitable enough for experiments of transformation in fungi. For every 105 protoplasts, about 15-25 protoplasts can regenerate after 24-36 h cultivation in a liquid medium and after 8-10 d in an agar medium. This study produced an efficient method for protoplast production, reverting them into a typical mycelia morphology using a Tulasnella calospora isolate.[返回]

论文摘要:Quantitative trait locus (QTL) detection was carried out for growth traits in 122 F1 progenies of Pinus elliottii var. elliottii (PEE) x p. caribaea vat. hondurensis (PCH) hybrid. The PCH male parent linkage map contained 108 markers in 16 linkage groups, while the PEE female parent contained 93 markers in 19 linkage groups. Sequence-related amplified polymorphism (SRAP), microsatellite (SSR), expressed sequence tag polymorphism (ESTP)and intetr-simple sequence repeat (ISSR) were selected from an existing linkage map. Growth traits investigated were height at age five(HT05) and six years (HT06), diameter at breast height at age five (DBH05) and six years (DBH06) and annual, growth from age five to six years of height (AGHT) and diameter at breast height (AGDBH), Kruskal-Wallis and interval mapping approaches were used to estimate levels of significance, the number of QTLs, the percentage of the phenotypic variation explained by each of QTLs and their positions on the genetic linkage maps. Twenty six QTLs with significance levels p < 0.05 were detected on the parental maps for the six growth tra ts,. wh ch exp a ned more than 15% of the phenotypic variation, suggesting an existence of major-effect genes. Several QTLs had the pleiotropy controlling more than one single growth trait. Overall, the proportion of phenotypic variation explained by QTLs ranged from 5.9% to 40.6% for HT05 and HT06 from 6.6% to 42.0% for DBH05 and DBH06 and from 5.8% to 22.3% for AGHT and AGDBH. The results from this study provide a basis for marker-aided selection (MAS) in PEE x PCH hybrids.[返回]

论文摘要:The present study on the sediment production in natural forests was carried out on an 13,284ha area, located in Guilan Province, in the north of lran. This area is Under intensive management. We first analyzed the physical factors of the area. Then, the amount of road construction, afforestation and harvested wood volume, as well as the harvesting method in the region were determined for the years between 1993 and 2010. The results show that the average annual amount of sediment production in the entire region, over this 18-year period, has increased annually at a rate of 3.54 t・km-2 (R2=0.056) at a confidence level of 95%, Operations management and management methods used in this forest area has affected the amount of sediment production. Clear cutting and extensive harvesting methods in the region caused an increase in the amount of sediment production, as indicated by a coefficient of detrmination of 0.71, impiying that forest management may be considered effective and in harmony with the environment.[返回]

论文摘要:Juniperus species (juniper) are resistant to drought and. have medicinal properties. In order to investigate the condition of juniper reproduction and prevent a decline in its area, largely caused by problems in natural regeneration, the asexual reproduction of juniper has received special attention besides its sexual propagation. Among the six species of juniper in Iran, the most abundant are Juniperus polycarpos and J. excelsa. Juniperus polycarpos is ambi-sexual and J. excelsa unisexual. We show. that collecting seeds: in September provided for better germination than in November. In sexual propagation 82.0% and in asexual (graft) 47.6% success were achieved. Compared to a mixture of water lime and hormones, a natural treatment of only water had better results. Diurnal change of temperature increased germination of juniper seeds over a constant temperature. Aphyllous stem cuttings from mature and old trees are iess successful in rooting than leafy cuttings of juniper saplings. A treatment with Aaterra significantly reduced the percentage of rooted cuttings and total root length, Aliette, Fongarid, Octave, Benomyl, Delsene M and Maneb fungicides Significantly increased the percentage 0f rooted Cuttings but did not affect total root length. Autumn explants formed more calli than spring explants. Successful multiplication was achieved by treating buds with a mixture of 0.7% polyvinyl pyrrolidone (PVP) and 2% sucrose and cultured in a basal Msor WRC medium with a maximum number of new buds. In consideration of extracted material of juniper species in remedy of infectious, fungus, Contagious diseases as well as for drought resistance and longevity of this species in severe .arid environmental Conditi0ns and Various industrial applications, reproduction and seed production by micro propagation and biotechnol0gy are necessary.[返回]

论文摘要:Five Populus x euramericana wood samples representing three different sites were selected and near linfrared (NIR) spectra were obtained. For these sections, basis weight, brightness and three mechan ca properties(tensile index tearing index and bursting index) were determined by standard analytical methods. Calibrations were developed for each paper property using the NIR spectra, data on paper properties, using partial least squares (PLS) regression. The results show that the coefficients of correlation of calibration and validation for basis weight were 0.8824 and 0.8299, respectively; the standard error of calibration. (SEC) and prediction (SEP) were 1150 and 1170, respectively. In testing for brightness the correlation coefficient of calibration was 0.9621 and for validation 0.9612, while the SEC and SEP were 0.997 and 1.300. respectively; paper brightness and NIR spectroscopy were highly correlated. NIR spectroscopy can be used to predict tensile, tearing and bursting indices of paper samples rapidly. We found, that the paper properties fitted by NIR and GB methods were highly correlated. The coefficients of correlation of calibration and validation for basis weight exceeded 0.8000, while the SEC and SEP were very small. These results reveal that the five paper properties of Populus x euramericana and those predicted by the NIR model were highly correlated. We conclude that the NIR models can be used for the prediction of paper properties.[返回]
