It is well known that nanomaterials are widely used in biomedicine as imaging tools, phototherapy agents, and gene delivery carriers. Here, we report the synthesis and characterization of novel amphiphilic polyurethane nanoparticles (APU NPs), and the interaction of the APU NPs with CT-DNA and cell-penetrating. The physical properties and chemical composition of APU NPs were characterized by transmission electron microscopy (TEM), dynamic light scattering (DLS), zeta potential and nuclear magnetic resonance (NMR). The interactions with CT-DNA were investigated by circular dichroism (CD) and ultraviolet-visible (UV-Vis) spectroscopies. The cell membranes penetration were characterized by fluorescence inverted microscope and HEK293 cell was used. These results showed that the APU NPs might cause slight conformational change in CT-DNA, and the APU NPs could efficiently internalized into the HEK293 cells without overt membrane disruption.In particular,50 nm of the nanoparticles can enter the cell by effectively penetrating cell membrane and no damage.
