超(亚)临界流体色谱拆分布洛芬对映体的热力学研究

摘要:本文以Kromasil CHI-TBB柱为手性固定相,在实验温度293~323K和压力12.0~21.0 MPa范围内用超(亚)临界流体色谱法拆分了布洛芬对映体,考察其热力学性质并与高效液相色谱法拆分相比较。计算了拆分过程中,对映体间焓变差值和熵变差值。在实验范围内,HPLC拆分时,容量因子的对数lnk’和分离因子的对数lnα均与1/T成简单的线性关系;SFC拆分时,lnk’与1/T不成线性关系而lnα与1/T成简单的线性关系。两种拆分都属于焓控过程,较低的温度有利于对映体的分离。

Thermodynamic study on the resolution of ibuprofen enantiomers by supercritical (or subcritical) fluid chromatography
Abstract:The resolution of ibuprofen enantiomers was studied on a chiral stationary phase of Kromasil CHI-TBB column. The thermodynamic properties of the enantiomers in supercritical (subcritical) fluid chromatography were investigated in the ranges of temperature 293~323K and pressure 12.0~21.0 MPa, and compared with those in high performance liquid chromatography. The differences of enthalpy changes and entropy changes for the transfer of enantiomers from the mobile phase to the stationary phase were calculated. The plots of lnk’ and lnα versus 1/T are straight lines in HPLC. The plots of lnα versus 1/T are still straight lines but the linear relationship of lnk’ versus 1/T could not be observed in SFC. The enthalpic contribution to the overall enantiomer transfer energy is generally more important than the entropic contribution in the temperature range examined, and lower temperature is better for chiral resolution of ibuprofen.
