体外诱导成年大鼠骨髓间质细胞向神经系细胞转化的试验研究

摘要:目的 验证并探讨成年大鼠骨髓间质细胞向神经系细胞定向诱导分化的可能性和方法。方法 利用成年大鼠骨髓培养骨髓间质细胞,经传代纯化后加入相应诱导剂,使其向神经元前体细胞及schwann细胞转化。观察诱导过程中细胞形态学变化,并利用免疫细胞化学染色的方法鉴定诱导细胞性质。结果 经过不同的诱导转化过程,骨髓间质细胞形态分别表现为多突触的神经元细胞样和呈漩涡样排列的双极细胞形态;免疫细胞化学染色结果显示,诱导后细胞分别对NSE,S-100,GFAP抗体呈阳性反应。结论 骨髓间质细胞在一定条件下可以向神经细胞系分化,有可能取代神经干细胞成为中枢神经疾病治疗及周围神经再生的种子细胞。

in Vitro Differentiation of Adult Rat Bone Marrow Stromal Cells into Cells of Neural Lineage
Abstract:Objective Verify and investigate the possibility and methods of adult rat bone marrow stromal cells’(MSCs) differentiation into cells of neural lineage. Methods After isolation and culture, MSCs were treated with different reagent and trophic factors sequentially to induce them to turn into cells with a phenotype similar to early progenitors of neural cells and schwann cells. Observing the morphologic change of MSCs, then identify them by immunocytochemistry methods. Results MSCs turned into neuron-like cells with process-like extensions and bipolar cells arranged into swirl after respective differentiation; Immunocytochemistry of the differentiated cells showed that NSE, S-100, GFAP stain positive. Conclusion Under certain conditions MSCs can differentiated into neural lineage cells, and may substitute neural stem cells as seed cells in central neural system disease therapy and peripheral nerve regeneration.
