物理技术与方法在蛋白结构与功能研究中的应用

摘要:现代物理技术与方法的发展为蛋白分子的研究提供了丰富而有效的手段。首先强调了技术与科学发展的辩证关系;介绍了在蛋白分子研究中的常用物理技术与方法如质谱技术、核磁共振技术、同步辐射X射线衍射技术、红外光谱和紫外光谱技术、差示扫描量热技术的物理原理及其在大分子研究中的应用现状,并重点介绍了一些新兴的非常规研究手段诸如原子力显微镜、激光光镊技术的特点及其在蛋白研究中的最新应用和研究成果;通过对比诸多技术与方法近年来在国内外研究中的进展情况,对各种技术与方法的优缺点进行了阐述;综合有效地运用这些仪器是必需的,但突出新技术与新方法更能代表这一学科发展的先进性。

Application of Physical Techniques
Abstract:he developing modern physical techniques offer a series of abundant and effective methods to the study on proteins in structure and function. Firstly stressed on the dialectic relationship between physical techniques and the improvement of science in history. And then introduced a lot of physical techniques in common use in proteins such as mass spectroscope, nuclear magnetic resonance, synchronization X-ray diffraction, infrared spectroscope, ultraviolet spectroscope and differential scanning thermal analysis, and reviewed their application status in quo to proteins. Secondly paid the attention to the burgeoning unconventionally instruments i.e. atomic force microscope and laser optical tweezers which have attracted the professional men in concern, and introduced latest application and researches’ fruits.
