自发性癫痫大鼠海马Ⅰ型与Ⅲ型钠通道α亚基表达上调

摘要:目的 探讨自发性癫痫大鼠(SER)海马Ⅰ型与Ⅲ型钠通道α亚基mRNA与蛋白的表达情况。方法 给大鼠测脑电图,发现癫痫大发作后即刻提取海马组织总RNA,应用逆转录-聚合酶链反应(RT-PCR)表达α1与α3亚基mRNA;应用免疫荧光定位与表达α1亚基蛋白与α3亚基ⅢN蛋白;应用免疫组化半定量表达α1亚基蛋白。结果 SER海马Ⅰ型与Ⅲ型钠通道α亚基mRNA表达显著高于WTC,(P〈0.01);SER海马Ⅰ型钠通道α亚基蛋白表达显著高于WTC (P<0.01);α3亚基ⅢN蛋白在SER有表达,而WTC无表达。结论 SER脑内海马Ⅰ型与Ⅲ型电压门控性钠通道α亚基表达上调,可能是神经元高兴奋性的原因或癫痫发作的继发表现。

The expression of typeⅠand Ⅲ sodium channelαsubunit was regulated up in spontaneously epileptic rat hippocampus
Abstract: OBJECTIVE Investigate mRNA and protein expression of typeⅠand Ⅲ voltage gated sodium channelαsubunit in spontaneously epileptic rat(SER) and wild typecontrol(WTC)hippocampus by control study. METHODS Total RNA were extracted from hippocampus after epilepsia gravior, and typeⅠand Ⅲ sodium channel α subunit mRNA expression were detected by RT-PCR respectively. TypeⅠand ⅢN sodium channel protein expression were detected by immunohistochemistry and immunofluoresence. RESULTS TypeⅠand Ⅲ sodium channel α subunit mRNA of SER expressed significantly higher than that of wild type control in hippocampus, (P<0.01). Type Ⅰsodium channel protein of SER increased significantly in hippocampus (P< 0.01), and ⅢN protein that should not have been expressed in adult rats expressed in adult SER hippocampus. CONCLUSION The expression of typeⅠand Ⅲ sodium channel α subunit was regulated up in spontaneously epileptic rat hippocampus,which may be the reason of neron hyperexcitability or succeeding appearance after seizure..
