Association of 45T/G Polymorphism in Adiponectin Gene and Plasma Adiponectin Level with Type 2 Diabetes

Abstract:The purpose of the present study was to investigate the association among SNP 45T/G and plasma adiponectin levels and the prevalence of type 2 diabetes(T2DM) in Uygurs of Xinjiang region, China. We performed a cross-sectional survey in representative sample of 151 Uygur adults aged 24-80 years. PCR-RFLP method was used to determine the distribution of allele and genotype frequency of SNP45 T/G polymorphism (exon2) in adiponectin gene. The enzyme-linked immunosorbent assay system (ELISA) was used to determine the plasma adiponectin levels. Logistic regression was used to screen risk factors of type 2 diabetes. Compared with the normal glucose tolerance (NGT) group, type 2 diabetic group displayed higher distribution of TG+GG genotype, G allele frequency and lower plasma adiponectin concentration of TG+GG genotype carriers than that of TT genotype. Compared with SNP45 T carriers in NGT group, the G carriers have higher level of SBP,DBP,LDL(P<0.05), CH(P<0.005), however in T2DM group, the G carriers have lower level of HOMA-ISI (P<0.05), and higher level of HOMA-IR (P<0.05). Adiponectin SNP 45 is positively correlated with prevalence of T2DM in Uygurs of Xinjiang. G allele carriers who possess a reduced plasma adiponectin concentrations may associate with insulin resistance.
