两种不同实验设计方法对人参皂苷提取工艺的比较研究

摘要:目的:通过比较星点设计—效应面法和正交试验设计法在人参皂苷提取工艺中的应用,考察星点设计-效应面法在中药有效成分提取工艺研究中的优越性和可行性。方法:以乙醇浓度,回流时间和溶剂量为自变量,人参总皂苷提取率为因变量对自变量各水平进行多元线性回归和二项式拟合,用效应面法进行较优化工艺条件的选择,并且与正交试验设计结果进行比较。结果:在皂苷提取率和皂苷含量相近的情况下,星点设计—效应面法确定的工艺条件中乙醇浓度可在70-80%选取,乙醇用量可在3-6倍间选取,提取时间最短可在60min内完成,而正交实验设计法的结果是90min,提取时间要多出30min。结论:星点设计—效应面法对最优工艺的选择不限于所设置水平,具有预测性,较传统的试验设计方法,结果更为精确,更接近客观实际,效果更好。

Study on the Extraction of Effective Constituents from Radix Polygalae
Abstract:Objective: Through compared central composite Design/Response Surface Methodology and Orthogonal Test to Application in Extraction of Radix Polygalae .Study on the Extraction of Effective Constituents from Radix Polygalae based on them. Method: Independent variables were ethanol concentration reflux time and solvent fold dependent variable was extraction rate of senegen in Radix Ginseng linear or nolinear mathematic models were used to estimate the relationship between independent and dependent variables Response surface methodology was used to optimize the process of extraction prediction was carried out through comparing the observed and predicted values and Compared with result of Orthogonal Test . Result:Under the situation similar to extraction rate of senegen and Glucoside content of saponin .In condition of process of central composite Design/Response Surface Methodology alcohol density 70~80% and alcohol dosage3~6 selected backstreaming can finished in 60min but it is 90min in Orthogonal Test .extract time spend more 30min . Conclusion: It shows that the central composite design/Response surface methodology is highly reasonable and snccessful.
