六种富勒烯衍生物对Taq DNA聚合酶的抑制作用

摘要:以HeLa细胞β-actin的聚合酶链式反应(polymerase chain reaction, PCR)分析为实验模型,研究了6种富勒烯衍生物对Taq DNA聚合酶抑制作用的差异。实验发现,6种富勒烯衍生物三丙二酸富勒烯、富勒醇、富勒烯丙二酸水解物、亚甲基富勒烯二膦酸四乙酯、亚甲基富勒烯二膦酸四乙酯的完全水解物和二丙二酸富勒烯对Taq DNA聚合酶存在不同程度的抑制作用。在本实验条件下,除了亚甲基富勒烯二膦酸四乙酯及其完全水解物以外,当4种化合物在体系中的终浓度为0.01 mM以上时,均不能检测到该PCR产物,尤其以二丙二酸富勒烯对Taq DNA聚合酶的抑制程度最强。结果提示富勒烯的生物学活性与其上的修饰基团有着很大的关系。

Inhibition of Taq DNA polymerase by six fullerene derivatives
Abstract:Inhibition of Taq DNA polymerase by six fullerene derivatives was investigated by using PCR analysis of β-actin in HeLa cells as an experimental model. It was discovered that trimalonic acid C60、fullerol、dimalonic[60]fullerene hydrolysate、tetraethyl methano[60]fullerenediphosphonate、methano[60]fullerenediphosphonic acid and dimalonic acid C60 exhibited different inhibitory effects on Taq DNA polymerase. Except tetraethyl methano[60]fullerenediphosphonate and methano[60]fullerenediphosphonic acid, another four [60]fullerene derivatives could inhibit Taq DNA polymerase at the concentration of 0.01 mM, especially dimalonic acid C60. The results revealed the relationship between biological activities of fullerene derivatives and their modified groups.
