水杨醛氨基酸Schiff碱铜配合物体外抗肿瘤作用

摘要:本文应用MTT比色法及流式细胞术研究四种水杨醛氨基酸Schiff碱铜配合物(6B、7B、6P和7P)对BGC823胃癌细胞系和HeLa宫颈癌细胞系的抑制生长和诱导凋亡作用。研究发现,不同种类的配合物对BGC823细胞和HeLa细胞的生长增殖均有明显的抑制作用,并且存在量效关系。四种配合物均促使细胞发生凋亡,凋亡率具有浓度依赖关系。同时也改变细胞周期时相分布,表现为G0/G1期细胞百分数显著减少, G1/S与G2/M期细胞百分数增加。

Effect of copper ternary coordination compounds of the salicylidene-amino acid Schiff base on cancer cell lines in vitro
Abstract:To study the anti-proliferation and inducing apoptosis effects of four copper ternary coordination compounds of the salicylidene-amino acid Schiff base(6B,7B,6P and 7P)on human gastric carcinoma cells BGC823 and human cervical carcinoma cells HeLa through MTT assay and the flow cytometry(FCM). Our study showed that the proliferation of BGC823 cells and HeLa cells were significantly inhibited by the four Schiff base complexes (6B,7B,6P and 7P)and the inhibition rates were concentration-dependent. FCM analysis showed the cells of drug treatment groups had higher apoptosis index(AI) and the effects were concentration-dependent. The phases of cell cycle were also changed: the percentage of cells in G0/G1 phase decreased, and the cells in G1/S and G2/M phase increased.
