金属离子对牛胰岛素淀粉样纤维形成的作用

摘要:蛋白质或者多肽转变为淀粉样纤维主要取决于所处的环境条件。本文采用ThT和ANS荧光、圆二色谱和偏振光显微镜等方法研究了溶液中金属离子(Mg2+, Cu2+, Zn2+, Co2+, K+, Li+ 和 La3+) 对牛胰岛素淀粉样纤维形成的影响,结果表明,所有金属离子对胰岛素淀粉样纤维化均有促进作用,其作用大小次序为:La3+ > M g2+ > Cu2+ > Zn2+ > Li+ > Co2+ > K+。这种作用与促进蛋白质疏水区暴露的作用是一致的。相关性分析显示,金属离子与蛋白质分子间的不同作用与其水合自由能和静电常数有关。并进一步由此结果推测了金属离子与蛋白质的作用机理,以及蛋白质淀粉样纤维化的分子机制。

he effects of metal ions on the amyloid fibrillation of bovine insulin
Abstract:The conversion of native polypeptide into amyloid fibril is strongly dependent on the physical and chemical environment. In the present study, we have examined the effects of metal ions on the amyloid fibrillation of bovine insulin. The growth kinetics and morphology of insulin fibril were monitored and characterized by thioflavin T fluorescence, ANS binding, circular dichroism and Congo Red assay. All metal ions tested herein accelerated the formation of insulin fibril with an order of La3+ > M g2+ > Cu2+ > Zn2+ > Li+ > Co2+ > K+. The growth of amyloid fibril was accompanied with a partially exposure of the hydrophobic interior of insulin. Correlation analyses showed the acceleration of insulin fibrillation was related to free energy of hydration and electrostatic contribution of the metal ions. Potential implications of these findings to the amyloid fibrillation of insulin are exploited and discussed.
