mGluR1在自发性癫痫大鼠海马中表达

摘要:目的 通过对照研究,检测自发性癫痫大鼠海马中mGluR1的表达情况。方法 通过RT-PCR技术检测鼠海马中mGluR1基因的表达;利用免疫组化和western blotting方法检测鼠海马中mGluR1总蛋白的表达。结果 自发性癫痫大鼠海马中mGluR1基因总水平下降(P〈0.05),免疫组化显示海马DG区和CA3区的mGluR1蛋白表达均下降(P〈0.05),CA1区无显著变化(P>0.05),尚存活的神经细胞mGluR1的表达较正常鼠增强;western blotting检测mGluR1蛋白总量下降(P〈0.05)。结论 自发性癫痫大鼠海马中mGluR1表达下调。

Expression of mGluR1 in hippocampus of spontaneously epileptic rats
Abstract:Objective Investigate the expression of mGluR1 in spontaneously epileptic rat hippocampus by control study. Methods Total RNA was extracted from hippocampus of SERs and wistar rats,and mGluR1 mRNA expressions were detected by RT-PCR respectively;The expression of mGluR1 protein were detected by immunohistochemistry and western blotting. Results mGluR1 mRNA of SER were lower than those of wistar rats in hippocampus(P <0.05).mGluR1 protein in DG and CA3 regions of SER hippocampus by immunohistochemistry were decreased compared with wistar rats(P <0.05),but in CA1 region had no significant difference (P﹥0.05).In survival cells, mGluR1 protein was enhanced. By western blotting,total mGluR1 protein was down-regulated in SER hippocampus(P <0.05). Conclusion The expression of mGluR1 was down-regulated in SER hippocampus.
