四种不同剂型匹罗卡品滴眼液在兔眼房水中的生物利用度比较

摘要:采用RP-HPLC法对匹罗卡品滴眼剂的四种不同剂型:1%溶液、1%的溶液脂质体混合液、1%脂质体和4%凝胶在兔眼房水中的药代动力学进行对比研究。120只兔均分为40组,每组3只兔(6只眼),每10组为一单位进行一种剂型的试验,每只兔眼滴入药物50 μL,分别测定给药后5、10、30、40、60、90、120、180、240及360 min时眼房水中匹罗卡品滞留量,用二氯甲烷提取房水中药物。结果显示,不同时间点1%的溶液脂质体混合液、1%脂质体和4%凝胶三种剂型滴眼剂的房水药物浓度均明显高于1%溶液滴眼液的房水浓度,尤以1%脂质体和4%凝胶最为明显,且1%脂质体滴眼剂在用药后6h,房水中仍可测出匹罗卡品。因此,将匹罗卡品滴眼液的剂型改为脂质体或凝胶可以明显提高匹罗卡品的生物利用度,增强疗效,减少用药频率,提高匹罗卡品对青光眼的治疗指数。

Evaluation of Ocular Bioavailability of Four Formulations of Pilocarpine Using the Albino Rabbit Eye Model by RP-HPLC
Abstract:In this paper, the ocular bioavailability for four formulations of pilocarpine was compared using the albino rabbit eye model in the aqueous humor by RP-HPLC method. The 120 albino rabbits were divided into 40 groups, each consisting of 3 animals. Every 10 groups received 50 μL of (A) 1% generic pilocarpine, (B) 1% mixture pilocarpine solution consisting of aqueous sample and liposome sample, (C) 1% liposome pilocarpine, (D) 4% gel pilocarpine, respectively. The aqueous humor was withdrawn at 5, 10, 30, 40, 60, 90, 120, 180, 240 and 360 min. Pilocarpine was extracted from aqueous humor with dichloromethane. The results showed that the pilocarpine formulations: B, C and D gave higher aqueous humor concentration of drug than formulation A at all time periods tested. The areas under curve of formulation B, C or D were several times as much as formulation A. Measurable amounts of drug following formulation B, C and D were still observed at 180, 360 and 240 min, respectively. Our study demonstrated the significantly increase in ocular bioavailability pilocarpine due to the therapy with liposome or gel. It shows that a novel topical formulation of pilocarpine in liposome or gel might serve as a long acting antiglaucoma agent, and less frequent applications are necessary with them than with commercial drops.
