DON毒素的提取、纯化

摘要:为了获得大量的、高纯度的脱氧雪腐镰刀菌烯醇(DON),选择来源于赤霉病小麦的禾谷镰刀菌进行培养,结果发现,禾谷镰刀菌在约含25%水的小麦培养基中25℃振荡培养,DON产量与培养天数呈近似抛物线关系,培养10天后,DON产量直线上升,30天达最高值,此后便迅速下降。收集培养基,经过在乙腈-水(84:16)溶液中振荡、过滤、石油醚脱脂、乙酸乙酯萃取、蒸发,并通过以无水硫酸钠加上弗罗里硅土为填料的层析柱提取纯化得到了高纯度的DON毒素,为该真菌毒素的深入研究提供了基础。

Isolation and Purification of Deoxynivalenol
Abstract:Fusarium graminearum was cultured in order to obtain a plenty of DON with high purity. The most suitable condition for Fusarium graminearum producing DON is that Fusarium graminearum was shaking cultured under 25℃ in wheat medium containing 25% water. The production of DON increased greatly after ten days and reached the top at thirty days, and dropped after that. The cultured wheat were collected and extracted with acetronile-water (84:16,V/V),and then purified by a chromatography column with Florisil and anhydrous sodium sulfate as fillings. At last, plenty of DON with high purity was obtained, which provided the basis for further researches on DON.
