PKCζ降低表达削弱了乳腺癌细胞的趋化运动和黏附能力

摘要:目的 研究蛋白激酶C ζ (PKCζ) 降低表达对乳腺癌细胞趋化运动和黏附能力的影响。 方法 利用RNA干扰技术特异性降低PKCζ蛋白在乳腺癌MDA-MB-231细胞中的表达,通过滤膜小室法 (Mircro-Boyden Chamber) 检测细胞的趋化运动能力,通过细胞黏附实验检测细胞的黏附能力。 结果 RNA干扰有效的降低了乳腺癌MDA-MB-231细胞中PKCζ蛋白的表达。在表皮生长因子受体 (EGFR) 诱导下,与参照细胞相比,PKCζ降低表达后的细胞趋化运动能力和黏附能力都显著下降。 结论 PKCζ降低表达削弱了乳腺癌细胞的趋化运动能力和黏附能力。

Reduction of PKCζ expression inhibits chemotaxis and adhesion of human breast cancer cells
Abstract:Objective The effect of PKCζ knockdown on the chemotaxis and adhesion ability of human breast cancer cells was investigated. Methods RNA interference was used to reduce the expression of PKCζ in human breast cancer MDA-MB-231 cells. The chemotaxis and adhesion ability of the cells were detected by Micro-Boyden Chamber assay and cell adhesion assay. Results The expression of PKCζ protein in MDA-MB-231 cells was effectively down-regulated by RNA interference. Both EGFR-induced cell chemotaxis and adhesion ability were inhibited. Conclusion Reduction of PKCζ expression impaired the chemotaxis and adhesion ability of human breast cancer cells.
