内皮祖细胞对肝硬化大鼠治疗的研究

摘要:目的 研究内皮祖细胞(EPCs)对肝硬化大鼠的治疗作用。方法 雄性大鼠提取EPCs,采用CCl4/橄榄油灌胃12w制成雌性SD大鼠肝硬化模型,随机取12w的大鼠20只分成2EPCs组 (n=10)和对照组(n=10)。在EPCs组,EPCs经门静脉回输入大鼠体内;对照组,门脉回输入等体积的生理盐水,移植4周后处死所有大鼠,取肝组织石蜡切片,进行H&E常规染色和Masson三色染色法;免疫组织化学染色检测α-SMA,Col III和Ki67,进行组织学和纤维化评估,同时取外周血进行生化检测评估肝功能。结果 与对照组相比,EPCs移植组中,HAI,ALT,AST和TBIL水平降低,Alb和Ki67水平高,α-SMA和Col III表达少(P<0.05);结论:移植EPC可以促进大鼠肝硬化的肝细胞增生,减轻肝纤维化程度。

The function of endothelial progenitor cells in rats with hepatic cirrhosis
Abstract:Objective To study the function of endothelial progenitor cells (EPCs) in rats with hepatic cirrhosis. Methods Hepatic cirrhosis rats were made by feeding with 25 % CCl4/olive oil for 12 weeks.12 weeks hepatic fibrosis rats were subdivided into EPCs group (n = 10) and saline control group (n = 10). EPCs were injected into the portal vein for 4 weeks in EPCs transplanted group. Rats in untransplanted group were killed at the beginning of 12 week. Rats in EPCs transplanted group and saline control group were killed at the beginning of 16 week. The clinical blood biochemical parameters from serum were examined. The degree of liver cirrhosis was evaluated by stains of Masson staining and by detecting the expression of α-SMA and Collagen III. Results Compared with saline control group, in EPCs group, hepatic activity index (HAI), the levels of ALT, AST and TBIL in serum were lower, the level of Alb and Ki67 was higher, the expression of α-SMA and Collagen III were lower(P<0.05). Conclusion EPCs infusion improve hepatocye regeneration and ameliorate established hepatic cirrhosis.
