蛋白激酶C与人视网膜色素上皮细胞吞噬功能关系的研究

摘要:目的 通过检测人视网膜色素上皮(HRPE)细胞特异性及非特异性吞噬过程中蛋白激酶C(PKC)活性的变化,明确PKC在HRPE的吞噬功能中所起的作用。方法 用1×107 个/ml视杆细胞外节膜盘(ROS)及乳胶微球(LB)于37℃孵育培养的正常HRPE细胞,在孵育的不同时间(5 min~48 h)终止吞噬反应。用双重荧光标记法检测HRPE细胞吞噬动力学。用扫描及透射电镜证明HRPE细胞对LB及ROS结合与吞噬。用液闪记数γ-32P放射活性法检测相应时间点的HRPE细胞特异性及非特异性吞噬时的PKC活性。 结果 HRPE细胞特异性吞噬过程中,孵育15 min时ROS结合于HRPE细胞表面;胞浆及胞膜PKC活性的改变发生在孵育5min时,但二者均在孵育24h达到最低值。在非特异性吞噬过程中,HRPE细胞结合LB发生在孵育90 min;胞浆及胞膜PKC活性在孵育的5min—48h时间内,与对照组比较均无明显差异(P>0.05)。结论PKC对HRPE细胞特异性吞噬吞入过程的维持十分重要,但不参与非特异性吞噬的直接调节。并且,HRPE细胞特异性吞噬过程伴随着PKC活性的降低。

The relationship between protein kinase C and the phagocytic fuction of human pigment epithelium cells
Abstract:Objective To investigate the role of protein kinase C (PKC) in the phagocytic process of human retinal pigment epithelium (HRPE) cells by measuring this two indexes in the specific and nonspecific phagocytic process. Method The cultured HRPE cells were incubated with 1×107 ml-1rod outer segments (ROS) or leatex beads(LB) at 37℃,then were rinsed the at different incubation time (5min-48h) to terminate the phagocysis.The kinetics of phagocytosis was measured by double- fluorescent vital assay.The binding and ingestion of ROS or LB were proved by scanning and transmission electron microscope. PKC activity was measured by counting γ-32P radio-activity with liquid scintillation. Result In specific phagocytosis of HRPE cells ,the binding of ROS happened at 15 min of incubation;the activity of PKC (both in cytoplasm and on membrane)decreased at 5 min,but both indexes reach their minimum level at 24h. In nonspecific phagocytosis of HRPE cells ,the binding of LB by HRPE cells happened at 90 min;on the other hand, when HRPE cells were incubated with LB, the activity of PKC(both in cytoplasm and on membrane) was stable during all incubation periods(p>0.05). Conclusion PKC are very important for sustaining the ingestion process in specific phagocytosis of HRPE cells, but has no direct realationship with nonspecific phagocytosis .Furthermore, PKC activity in HRPE cells decreases during specific phagocytosis.
