针刺对2型糖尿病大鼠肝组织IRS-2mRNA的影响

摘要:目的:探讨大鼠肝组织胰岛素受体底物2(IRS-2)基因表达在2型糖尿病(T2DM)发病机制中的作用及针刺对T2DM大鼠肝组织IRS-2基因表达的影响。方法:SD雄性大鼠100只,经高脂饲料喂养成肥胖大鼠(50只),腹腔注射小剂量链脲佐菌素(Streptozotocin,STZ)(25mg/kg)造模成T2DM大鼠(18只),随机分为针刺组、药物组(优降糖)和模型组,处理4周后,用快速血糖仪检测空腹血糖(FBS)、用放免法检测空腹胰岛素(FINS)、用实时定量荧光PCR(real-time RT PCR)法检测肝组织IRS-2的基因表达,并与正常组大鼠进行对照。结果:针刺组和药物组FBS水平比模型组显著降低(P<0.05),接近正常组水平(P>0.05);针刺组FINS明显低于模型组(P<0.05),接近正常组水平(P>0.05),药物组FINS水平亦低于模型组,但无显著差异(P>0.05);模型组IRS-2mRNA相对含量为正常组的2.19倍,针刺组IRS-2mRNA相对含量为正常组的4.84%,是模型组的2.21%,药物组IRS-2mRNA相对含量为正常组的4.59倍,是模型组的2.10倍。结论:肝组织IRS-2mRNA表达异常可能是T2DM的发病机制之一,针刺具有显著抑制T2DM大鼠肝组织IRS-2mRNA表达的作用。

Effects of Acupuncture on Hepatic IRS-2mRNA Expression in Type 2 Diabetes Mellitus Model Rats
Abstract:Objective: To research the effect of rat’s hepatic IRS-2mRNA on pathologic mechanism of type 2 diabetes mellitus and the effect of acupuncture on the expression of hepatic IRS-2mRNA in T2DM model rats . Method: 50 of 100 SD male rats fed with high fat feedstuff became fat rats, which were injected in abdomen with Streptozotocin(25mg/kg) to make T2DM rat models. Then T2DM rats(18) were Stochastically devided into Glyburide group(6), acupuncture group(6) and model group(6). After treatment for 4 weeks, fast blood sugar(FBS) was examined by active blood sugar meter and fast insulin(FINS) was examined by radioimmunoassav(RIA) and expression of hepatic IRS-2mRNA was examined by real-time RT PCR, the results were compared with normal rats. Result: After treatment, the level of FBS in acupuncture and Glyburide group are obviously lower than model group(P<0.05); the level of FINS in acupuncture is markedly lower than model group(P<0.05), the level of FINS in Glyburide group is also lower than model group but there has no statistical meaning between them(P>0.05). The IRS-2mRNA of model group is 2.19-fold of normal group; which of Glyburide group is 4.59-fold of the normal group ; which of acupuncture group is 4.04% of the normal group. Conclusion: The abnormal expression of hepatic IRS-2mRNA may be one of the pathologic mechanism of T2DM. Acupuncture significantly depress the expression of hepatic IRS-2mRNA in T2DM rats.
