洋葱核仁的超微结构动态变化特征和rRNA转录

摘要:以洋葱为材料对细胞周期中核仁的结构的动态变化进行了追踪,发现在常规电镜下S期的核仁的表面与G1、G2相比出现了大量毛状突起并通过毛状突起加强了与核仁周边染色质的联系,这种现象持续到S与G2的过渡时期,结合NAMA-Ur DNA特异性染色,柠檬酸铽RNA特异性染色,对这种现象进行了分析,认为这种核仁表面出现大量毛状突起现象是DNA进行复制期间,核仁在常规电镜下的一个明显特征。柠檬酸铽RNA特异性染色显示单链RNA分布于DFC及GC区,FC中未见有分布,说明DFC是rRNA进行转录的区域。

Dynamic change of the nucleolar ultrastructure and rRNA transcription within Allium cepa nucleolus
Abstract:In this paper, the ultrastructure in different phase nucleoli of Allium cepa were observed with the conventional electron microscopy. We found the nucleolus rim showed hairy and thus more linking of the nucleolus rim to the peripheral chromatins in S phase. By using the NAMA-Ur DNA specific staining technique and terbium cirate single-stranded RNA (ssRNA) special staining, we investigated the morphological changes of the nucleolus. The results showed this kind of phenomenon was the obvious characteristics of nucleolar structure during DNA replication. Further observations demonstrated that ssRNA mainly distributed in DFCs and their peripheral regions. It therefore can be concluded that the DFC, and not the FC, is the site of rRNA transcription in nucleoli.
