摘要:目的:研究吲哚美辛干预与NGX6基因对HT-29结肠癌细胞生物学行为的改变以及产生的分子机制。方法:通过MTT比色法检测细胞生存率、AO/EB染色结合荧光显微镜技术和双色荧光流式细胞仪检测细胞凋亡、体外侵袭转移实验和划痕标记染料示踪实验检测间隙连接通讯能力。结果:NGX6基因与吲哚美辛协同抑制HT-29细胞的增殖,降低HT-29细胞的转移能力,协同恢复间隙连接细胞间通讯能力;但对促进细胞凋亡和抑制细胞外基质粘附无协同作用。结论:NGX6基因与吲哚美辛协同抑制HT-29细胞的增殖和降低HT-29细胞的侵袭转移能力的作用

Synergistic effect of Indomethacin and NGX6 on the proliferation and invasion of human Colorectal Cancer Cells
Abstract:Object This study was designed to investigate whether indomethacin has synergistically inhibitory effect with NGX6 on the growth and invasiveness of human colon cancer cells (HT- 29). Method The cell proliferation was assessed by MTT assay. Cell apoptosis was assessed by Acridine Orange/Ethidium Brimide Staining (AO/EB), and Annexin-V-FITC/PI assay. Invasive behaviors of colorectal cancer cells were examined by cell adhesion, migration, and invasion assay. Gap junctional intercellular communication (GJIC) was assessed by the scrape-loading/dye transfer technique. Results The studies indicated that indomethacin and NGX6 had the synergistic effect on inhibition of the proliferation and invasion of HT-29 colon cancer cells, restoration of the gap junctional intercellular communication of HT- 29However, the effects on the enhancement of apoptosis and on the suppression of extracellular matrix adhesion of HT- 29 cells were not synergistic. Conclusion: Indomethacin and NGX6 gene inhibit the proliferation and invasion of HT-29 colon cancer cells.
