一个可以用来检测小麦背景中簇毛麦遗传物质的分子标记

摘要:一年生二倍体簇毛麦(Dasypyrum villosum)和多年生四倍体簇毛麦(D. breviaristatum)在异染色质的分布上具有差异性。从多年生簇毛麦基因组中分离的类Sabrina长末端反转录转座子序列OPH121182可以作为分子标记用来检测多年生簇毛麦染色质。本研究确证该序列在二倍体簇毛麦染色体组中的分布与应用价值。用根据OPH121182设计的引物 H12F和H12R对含二倍体染色质的物种以及近年来四川省区试材料进行扩增,结果显示OPH121182存在于不同居群的二倍体簇毛麦、硬粒小麦-簇毛麦双二倍体、6V(6D)、6VS/6AL、育成品种蜀麦375和内麦9号等材料中,这表明该标记也可以用于检测小麦背景中的二倍体簇毛麦染色质。PCR和荧光原位杂交(FISH)结果均显示该标记分布于簇毛麦全基因组,因此,分子标记OPH121182可以广泛用于小麦与簇毛麦属物种的远缘杂交育种工作中。

A novel molecular marker for the detection of Dasypyrum chromatin in wheat background
Abstract:In order to develop the molecular marker universally applied to the detection of the chromatin from Dasypyrum, which consists of annual Dasypyrum villosum and perennial D. breviaristatum. Studies showed that a Sabrina-like Long Terminal Repeat (LTR) retrotransposon sequence OPH121182 from D. breviaristatum genome could be used as molecular marker for detecting of D. breviaristatum and its derivatives. The aim of this study is to corroborate the feasibility of detecting the D. villosum chromatin and its derivatives using this marker. D. villosum accessions and wheat varieties or lines newly developed in Sichuan Province were tested using primer pair H12F and H12R, which was designed according to the sequence of OPH121182, the result showed that OPH121182 amplified in D. villosum accessions, Triticum- D. villosum amphiploid, 6V (6D) substitution line, 6VS/6AL translocation line, Shumai-375 and other several wheat lines, indicating that the marker could also be used for detection of D. villosum chromatin in wheat background. Furthermore, we also use the fluorescence in situ hybridization (FISH) of OPH121182 to identify the wheat - D. breviaristatum introgressions. It is concluded that OPH121182 was an effective molecular marker which can be widely used in chromosomal manipulation of wheat breeding using Dasypyrum species.
