DNA甲基化检测方法的研究进展

摘要:随着分子生物学新技术的不断突破,生物医学等相关领域得到了长足的发展,DNA甲基化的研究逐渐成为了近年来的热点,各种各样的甲基化检测方法被相继发展起来以满足不同类型的研究和临床的要求。根据不同检测方法的原理及其发展先后,大致可分为两大类: (1) 用甲基化敏感的限制性内切酶使DNA部分降解,再区分甲基化与非甲基化的相关序列; (2) 利用重亚硫酸盐(Bisulfite) 预处理DNA, 直接通过引物延伸反应或耦联其他机制对甲基化与非甲基化的胞嘧啶进行区分。我们依据这一分类,结合本实验室在DNA甲基化检测方法方面的研究特点,对目前常用的甲基化检测方法作一综述。

Summarization of technique for DNA methylation detection
Abstract:Biomedical and its related fields have been gotten great progress with the development of novel technology in life sciences. The studies of DNA methylation in recent years become the hotspot, and variety of methods has been developed to meet the different types of research and clinical requirements. According to different detection pattern and the principle, the method can be broadly classified into two categories: (1) methylation-sensitive restriction enzyme based detection method, the differences between methylation and non-methylation can be distinguished by the its sequences (2) converting the sample DNA by using heavy sulfite (Bisulfite), and the differences of methylation and non-methylation can be distinguished directly through the primer extension reaction or coupled with the non-methylation of cytosine methylation of distinction. Based on this classification, we summarized the method of DNA methylation combined with our own researches about DNA mthylation detection in the laboratory.
