部分国内外小麦种质材料HMW-GS分析

摘要:为了给小麦品质遗传改良中亲本的选配提供参考依据,本研究利用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)对 135 个国内外引进的种质材料和 973 课题中的 100 个 小麦骨干亲本及其衍生材料的高分子量谷蛋白亚基组成(HMW-GS)进行了分析。结果表明,235 份材料中,Glu-A1 位点编码的HMW-GS只有亚基 1 和 Null 两种变异类型,其中 Null 类型所占比例较高(78.65%),1 亚基较少(21.28%);Glu-B1 位点编码的HMW-GS有 8 种变异类型,亚基 7+8 和7+9 所占比例较高分别为 45.53% 和 32.34%。而优质亚基 14+15,14+15、13+16 和17+18 较少,分别为 7.23%、2.55% 和1.70%;Glu-D1位点编码的HMW-GS变异类型主要是亚基 2+12 和 5+10亚基。频率分别为74.89% 和 24。64%;共检测到 24 种 HMW-GS组合类型,其中亚基组合(N,7+8,2+12),(N,7+9,2+12)类型最多,分别占32.76% 和 22.13%,总材料中三位点具有优质亚基组合的频率较低(7.66%),在国内外引进的种质材料中的频率为 11.85%,973 材料的频率仅有 2%。在小麦品质改良中应注重部分国内外引进的种质材料的利用,部分 973 骨干亲本及其衍生材料可作为优良亲本质源。

Analysis on HMW-GS Composition of some Wheat Germplasms from China and Overseas
Abstract:The Composition of High-Molecular-Weight Glutenin Subunits(HMW-GS)of 135 Domestic and Overseas Wheat Germplasms, 100-wheat backbone of the material and its derivatives in 973 subjects were analyzed by SDS-PAGE method in order to provide basis of parents use in quality hereditary improvement. The results showed that in the 235 wheat materials, only two kinds of HMW-GS on Glu-A1 were examined, which are Null, 1 and the frequency of Null was higher(78.72%),1 was less(21.28%);There were eight kinds of HMW-GS on Glu-B1, In them, the frequency of 7+8 and 7+9 was 45.53 % and 32.34 % respectively, which was more higher than the other types. the frequency of quality subunit 14 +15,13 +16 and 17 +18 was 7.23%, 2.55%, 1.70% respectively, which was less.; about Glu-D1 , 2 +12 and 5 +10 appears more frequently, the frequency was 74.89%, 24.64% respectively. And 24 kinds of HMW-GS combination of diffrent types were examined. In them, the frequency of(N,7+8,2+12),(N,7+9,2+12)was 32.76 %,22.13%respectively, which was more higher than the other types.In the 235 wheat materials,7.66%with combination of three high-quality subunits was examined.in Domestic andOverseas Wheat Germplasms, the frequency of combination of three high-quality subunits is 11.85%,but the frequency is only 2% in wheat backbone of the material and its derivatives in 973 subjects. in quality hereditary improvement ,we shoud pay attention to Domestic and Overseas Wheat Germplasms, and the wheat backbone of the material and its derivatives in 973 subjects may be as sources of good-nature.
