一个新精原干细胞系的建立、冻存、移植与后代产生

摘要:精原干细胞具有自我更新和分化为成熟精子的能力。精原干细胞维持一定的数目以及保持正常的增殖、分化功能是确保男性生育的关键。正是由于精原干细胞的这些重要性,近年来越来越多的研究投入这个领域之中。在本研究中,我们第一次使用来源于成年C57小鼠精原干细胞未经任何遗传修饰建立了一个新的细胞系。该细胞系表现为Thy-1+, Oct4+, SSEA-1-, C-kit-细胞亚群。经过长期维持培养和冻存、复苏,该细胞系在移植入经白消安处理的受体小鼠后能够产生正常后代。

Generation of mice by transplantation of a novel functional spermatogonial stem cell line after cryopreservation
Abstract:Spermatogonial stem cells (SSCs) have the dual capacity of self-renewal and differentiation into mature sperm. Retaining adequate number as well as normal proliferation and differentiation of SSCs is the key to male fertility. Therefore, there is growing interest in studies of SSC. In this study, we for the first time established a novel SSC line derived from SSCs of adult C57 mouse without any genetic modifications. This cell line represented Thy-1+Oct4+SSEA-1-c-kit- subpopulation. After long term culture and cryropreservation, these cells were transplanted into busulphan-sterilized recipient mice, which produced normal offspring.
