加压毛细管电色谱分离盐酸奥昔布宁对映体

摘要:建立盐酸奥昔布宁加压毛细管电色谱(pCEC)的分离方法,采用羟丙基-β-环糊精(HP-β-CD)作为手性选择剂,在优化条件下盐酸奥昔布宁对映体达到了基线分离。考察了手性选择剂浓度、流动相配比、背景电解质pH值、柱温和分离电压对分离的影响,HP-β-CD浓度为16g•L-1,流动相配比水:乙腈(V/V)40:60、背景电解质pH2.8、柱温为18℃、分离电压20kV。采用毛细管电色谱方法分离盐酸奥昔布宁对映体比手性固定相法经济、比高效液相手性添加剂法快捷,本方法操作简单快捷,结果准确,可用于盐酸奥昔布宁对映体的分离,为开展此类药物对映体的研制和临床研究提供一种有效可靠的分析方法。

Enantiomeric separation of Oxybutynin chloride by Pressurized Capillary electrochromatography
Abstract:To establish a method to separate oxybutynin chloride enantiomers by pressurized capillary electrochromatography (pCEC) ,using the hydroxypropyl -β-cyclodextrin(HP-β-CD)as chiral selector. The best experimental conditions that mexiletine hydrochloride enantiomers were separated by baseline were obtained by optimizing the main experimental conditions: such as hydroxypropyl-β-cyclodextrin concentration, background buffer pH, capillary temperature, voltage and the mobile phase consisted,hydroxypropyl-β-cyclodextrin concentration 16g•L-1, the mobile phase consisted water:ACN (V/V)40:60, background buffer pH 2.8, capillary temperature 18℃, voltage 20kV . Oxybutynin chloride enantiomers were separated under the best experimental conditions by baseline which was more economically and rapidly than chiral stationary phase、high performance liquid chromatographic separation of oxybutyn in enantiomers using chiral mobile phase additive.The method is simple,rapid,and can be used for the separation of oxybutynin chloride enantiomers with satisfactory accuracy. It can provide an effective and reliable analysis method for the clinical research and development of the drug.
