家蚕热休克蛋白基因Bmhsp19.9启动子的活性分析

摘要:通过PCR扩增出家蚕(Bombyx mori)热休克蛋白基因Bmhsp19.9启动子的序列,TA克隆后进行序列测定,并构建重组载体进行瞬时表达。结果显示,该序列长为1 258 bp,具有典型的热激蛋白启动子的结构特点;序列分析显示,hsp19.9启动子区域可形成潜在的复杂茎环结构,推测其与启动子的功能有关。瞬时表达试验结果显示,hsp19.9的启动子能通过热诱导在家蚕BmN细胞和家蚕组织中驱动红色荧光蛋白报告基因(DsRed)瞬时表达,表明所克隆的hsp19.9启动子序列具有热休克蛋白基因的启动子活性。

Activity Analysis of Promoter of Bmhsp19.9 Gene from Bombyx mori
Abstract:The promoter of bmhsp19.9 sequenced after cloned by PCR and TA clone was cloned into a recombinant transient expression vector. The results showed the promoter was 1 258 bp and harbored typical structural characteristics of HSP promoters. Sequence analysis showed that the promoter region could form potential complicated stem-loop structures which may relate to the promoter function.The result of transient expression showed that the promoter of Bmhsp19.9 gene could induce transient expression of DsRed gene in BmN cells and silkworm tissues, which indicated the cloned Bmhsp19.9 promoter sequence possessed HSP promoter activity.
