一株葫芦内生真菌代谢产物的分离研究

摘要:本文以抑制枯草芽孢杆菌活性为筛选模型,自葫芦果实内筛选得到一株内生真菌。将该菌株发酵液进行固液分离,采用20%、50%、70%乙醇分级萃取菌体沉淀,经旋转蒸发、真空冷冻干燥除去大量溶剂后,制备得抑菌活性物质浓缩液,结果显示该物质的极性与氯仿相近,且对热较稳定。经氯仿二级萃取等操作,制备抑菌活性物质的固体粗产物,该产物经薄层层析后,显示1个具有抑菌活性的层析斑。最后对具有抑菌活性的层析斑进行HPLC分析,结果显示该层析斑中含有6个组分。

The separation of antibiotic substances of endophype fungus from calabash fruit
Abstract:A strain of endophype fungus was isolated from the calabash fruit with Bacillus subtilis as indicator bacteria. The mycellium precipitation were isolated from fermented broth by centrifugation, and then was extracted gradually by 20%, 50%, 70% aqueous solution of ethonal. The concentrated solution of the antibiotic substances was achieved by rotation evaporation and freezing desiccation. The antibiotic substances show similar polarity to that of chloroform and good thermal stability. The crude product of antibacterial substances were obtained by two-step extraction with chloroform and then analyzed by thin-layer chromatography. The chromatographic spot of the bioactive substance was further purified by HPLC giving rise to 6 components.
