MAE−HPLC−FD法同时测定粉葛根中葛根素和大豆苷元

摘要:本文采用响应面分析法(RSM)对微波提取条件进行了优化,建立了微波辅助提取 (MAE)-高效液相色谱(HPLC)-荧光检测法(FD) 同时测定粉葛根中葛根素和大豆苷元含量的方法。结果表明,测定葛根素和大豆苷元的线性范围分别为5.0 - 1000 mg/L和1.0 - 200 mg/L,平均回收率在93.8% - 109.6%之间,RSD < 7.4%。和药典方法比较,此法简便、准确、快速并且有更高的提取率。此法可用于药材中葛根素和大豆苷元的测定,也可用于含这两种异黄酮的药材的质量评价。

MAE-HPLC-FD for simultaneous determination of puerarin and daidzein in Radix Puerariae thomsonii
Abstract:A HPLC method was developed for simultaneous determination of puerarin and daidzein in Radix Puerariae thomsonii based on microwave-assisted extraction (MAE) and fluorescence detection (FD). Optimal extraction parameters were obtained through systematic investigation with response surface methodology (RSM) and chromatographic conditions were optimized. The method presented satisfactory precision (RSD <7.4%), good recovery (93.8–109.6%) and good linear relations (5.0–1000 µg/mL for puerarin and 1.0–200 µg/mL for daidzein). Compared with a method from the Chinese Pharmacopoeia, this method enabled not only simultaneous determination of puerarin and daidzein in Radix Puerariae thomsonii, but was simpler, more rapid and more accurate, with higher extraction efficiency. It should be of potential value in quality assessment of Radix Puerariae thomsonii medicinal materials.
