超低温保存日本牡丹花粉生活力测定

摘要:采用悬滴法在5%蔗糖+0.1%硼酸的培养液上对23个日本牡丹品种新鲜花粉及超低温保存1年后的花粉萌发率进行了测定和比较。结果表明,不同牡丹品种新鲜花粉生活力均存在差异,23个品种中,‘大藤锦’花粉萌发率最高,达到60.89%,而‘岛大臣’最低,为3.49%;超低温保存芍药花粉的效果因品种而异,但绝大部分品种花粉保存效果良好。

Pollen Cryopreservation of Japanese Tree peony
Abstract:With 23cultivars of Japanese Tree peony (Paeonia suffruticosa),pollen germination rate before and after cryopreservation were tested by hanging drop method. The culture medium is 5% sucrose+0.1% H3BO3. Results indicated that fresh pollen viability of different cultivars were obviously different,the germination rate of ‘Da Tengjin’ reached 60.89%, while it was only 3.49% for ‘Dao Dachen’. The germination rates of pollen stored in liquid nitrogen varied with cultivars and most pollen kept high viability after cryopreservation.
