川芎嗪对大鼠局灶性脑缺血后空间认知障碍和SVZ

摘要:目的:本研究旨在探讨TMP对大鼠局灶性脑缺血后空间认知障碍的改善作用以及对脑缺血诱导的SVZ神经发生的影响。方法:采用线栓法制作大鼠局灶性脑缺血模型,腹腔注射TMP,Morris水迷宫方法检测大鼠空间学习记忆功能变化,BrdU掺入法检测SVZ增殖的细胞。结果:本研究结果显示,大鼠局灶性脑缺血可造成严重的学习记忆功能缺陷,并可诱导SVZ的细胞增殖。Morris水迷宫实验中,TMP治疗后的大鼠游泳逃避潜伏期较缺血组显著缩短(P < 0. 05),大鼠穿经平台次数和在平台象限停留时间均较缺血组延长。TMP组SVZ的BrdU阳性细胞数在缺血后3和7d较缺血组显著增多。结论:TMP治疗可减轻脑缺血后的学习记忆功能障碍,促进缺血后SVZ的神经发生。提示TMP可能在成年脑损伤后的神经发生和功能恢复过程中发挥着一定的促进作用。

Effect of tetramethylpyrazine on spatial cognition and cell proliferation of SVZ after focal cerebral ischemia in rat
Abstract:Object: In the study, we aim to demonstrate the effect of tetramethylpyrazine (TMP) on spatial learning and memory, and if TMP can induce cell proliferation of SVZ after focal cerebral ischemia in rat. Methods: The focal cerebral ischemic model of rat was made by suture occlusing the middle cerebral artery. TMP was administered introperitoneally after MCAO. The spatial learning and memory function was studied by Morris water maze test. The cell proliferation of SVZ was labelled with BrdU and BrdU positive cells were showed by Immunohistochemical stain. Results: The ischemia group exhibited seriously spatial learning and memory deficits. There was a significant increase of BrdU positive cells in SVZ after MCAO. In place navigation trial, the mean values of escape latency of TMP group were shorter than those of ischemia group (P<0.05). In sp atial probe trial , there we re significant difference in the time percentage of spending in the former platform quadrant and frequency of crossing the former platform site between TMP group and ischemia group (P < 0. 05) . There was a significant increase of BrdU positive cells in SVZ of TMP groupcompared to ischemia group (p<0.05) at 3d and 7d. Conclusion: TMP can improve the spatial congnition defects induced by the focal cerebral ischemia, and promote cell proliferation in SVZ.
