肝组织工程血管流道结构的仿生设计

摘要:组织工程技术有望实现肝脏的工程化制造以解决肝移植手术中的供需矛盾,复杂的血管流道系统的设计与成型为目前组织工程的瓶颈技术。本文通过研究动物肝脏铸型结构,测量、统计了大鼠血管的特征数据,并利用结构约束优化算法对血管模型树进行仿生设计,进而利用voronoi图对产生的模型树进行评估与改进,构建出的计算机模型可直接用于制造肝组织工程支架的快速制造。

Biomimetic Modeling and Fabrication of Vascular System for Liver Tissue Engineering Scaffold
Abstract:Biomimetic vascular network system is crucial to engineer realistic liver equivalents in vitro. In this study, the vascular cast of rat liver was statistically analyzed and the key parameters such as branching angle were measured. The biomimetic vascular system for liver tissue engineering scaffold was modeled based on fluidic dynamics and constrained constructive optimization methods. The uniformity of vascular terminals in a given perfusion region was investigated using Voronoi technique. The results show that the final vascular model has the similar mean branching angle. Furthermore, this method can be further integrated with CAD second-development technique to present solid vascular model for rapid prototyping fabrication. The novel vascular system could facilitate the deliver of nutrients and oxygen in three-dimensional scaffold, which provides a new way to construct complex liver architectures for tissue engineering applications.
