加味温胆汤对抑郁模型大鼠海马显微结构及PKA、CREB mRNA表达的影响

摘要:目的:探讨加味温胆汤对抑郁模型大鼠海马形态结构及PKA-CREB信号转导通路的影响。方法:40只SD大鼠随机分为正常组、模型组、百忧解组及加味温胆汤组,应用孤养加慢性轻度不可预见性应激方法复制大鼠抑郁症模型,采用HE染色法标记各组大鼠海马CA3区神经元,原位杂交法检测各组大鼠海马CA3区PKA、CREB mRNA表达,病理图像分析系统分析比较各组差异性。结果:模型组大鼠海马CA3区神经元明显受损,与正常组比较,神经元计数显著下降(P<0.05),PKA、CREB mRNA表达明显降低(P<0.05);与模型组比较,中、西药组海马CA3区神经元受损明显减轻,神经元计数明显增加(P<0.05),PKA、CREB mRNA表达显著增强(P<0.05)。结论:加味温胆汤的抗抑郁作用可能与其活化海马区PKA- CREB信号通路、拮抗海马神经元损伤有关。

The Effects of Jia Wei Wen Dan Decoction on Depression Model Rats’ Microstructure and PKA/CREB mRNA expression of the Hippocampus
Abstract:Aim: TO investigate the effects of Jia Wei Wen Dan decoction (WD) on the hippocampus morphology and PKA-CREB signaling pathway of depression model rats. Methods: 40 rats were randomly divided into normal group/ model group/ fluoxetine group and WD group; reproduce the models of depression with the method of living lonely and unpredictability of chronic mild stress. Marked the neuron in the CA3 region of the hippocampus by HE staining, detected the expression of PKA/CREB mRNA in the CA3 region of the hippocampus by situ hybridization, and then compared the difference between them by using the pathologic imagine analysis system. Results: The neuron in the CA3 region of the hippocampus was damaged obviously, compared with normal group, the neurons count reduced significantly (P<0.05), and the expression of PKA/CREB mRNA were markedly reduced too(P<0.05); The damage of neuron in fluoxetine group and WD group relieved, and the neurons count went up markedly, the expression of PKA/CREB mRNA increased significantly(P<0.05). Conclusion: The mechanism of Jia Wei Wen Dan decoction on treating depression is related to active the PKA-CREB signaling pathway and to protect the neuron in the CA3 region of the hippocampus from hurting.
