缺氧对体外培养的大鼠胚胎皮质神经干细胞增殖的影响

摘要:本实验采用细胞培养和免疫细胞化学等方法,研究缺氧条件对大鼠脑神经干细胞(NSCs)增殖作用的影响。实验结果显示,应用MTT比色分析,与对照组比较,缺氧后增殖细胞数量显著增多(P<0.05),其增殖作用与EPO对培养NSCs增殖的刺激作用相似。神经球直径测量表明,缺氧组神经球直径比对照组显著增大。用流式细胞仪检测,缺氧后分裂期细胞的比例明显增加。研究结果提示,缺氧可明显刺激培养的大鼠脑神经干细胞的增殖作用,对研究脑缺血修复的机制具有参考价值。

Influence of hypoxia on cell proliferation of NSCs derived from embryonic rats cortex in vitro
Abstract:Influence of hypoxia on cell proliferation of rats NSCs was investigated by means of cell culture, immunohistochemistry and so on. After hypoxia, the MTT assay showed that the Proliferated NSCs increased significantly compared with control ( P<0.05 ), which resembled that NSCs proliferation was stimulated by EPO. Diameter measure demonstrated that, after hypoxia treatment, diameter of neurospheres increased compared to normoxic cultured neurospheres. Cell cycle analysis showed PI and SPF were significantly increased induced by hypoxia. Our study show that hypoxia can obviously induce the NSCs proliferation in vitro, which is reference to study of cerebral ischemic repair.
