抗冻剂对鮸鱼精子超低温冻存效果的影响

摘要:研究了Gly、DMSO、EG及PG四种抗冻剂对鮸鱼精子超低温冻存效果的影响。结果表明,以Cortland溶液为稀释液,10%、15%浓度的Gly,5%、10%、15%浓度的DMSO,10%浓度的EG,以及10%、15%、20%浓度的PG为抗冻剂超低温冻存鮸鱼精子的效果较好,冻精活力与鲜精接近,其中,以10%Gly为抗冻剂冷冻保存的效果最佳,冻精激活率、运动时间及寿命分别达86.38%±1.63%、8.24min±1.37min、10.21min±0.42min;而以5%、20%、25%、30%Gly,20%、25%、30%DMSO,5%、15%、20%、25%、30%EG,以及5%、25%、30%PG为抗冻剂超低温冻存鮸鱼精子的效果较差,冻精活力与鲜精差异显著。初步认为,鮸鱼精子超低温冻存用抗冻剂宜选用10%Gly、或10%DMSO、或10%EG及15%PG。

Effects of Cryoprotectants on Miichthys miiuy Sperm Cryopreservation
Abstract:The effects of four cryoprotectants on on Miichthys miiuy Sperm Cryopreservation were investigated. The four cryoprotectants were Gly, DMSO, EG and PG. The results indicated that the vitality of frozen -thawed sperm were close to fresh sperm while Cortland solution was used as extender and 10%, 15%Gly; 10%, 15%DMSO; 10%EG; 10%, 15% and 20%PG were used as cryprotectants. The best activation rate,moving time and survival time of frozen-thawed sperm were 86.38%±1.63%, 8.24min±1.37 min and 10.21 min±0.42 min in which 10% Gly was used as cryoprotectant for cryopreservation. However, there were significant differences between fresh sperm and frozen semen which preserved with 5%, 20%, 25%, 30%Gly; 20%, 25%, 30%DMSO; 5%, 15%, 20%, 25%, 30%EG and 5%, 25%, 30%PG. It was suggested that the optimal cryoprotectants for Miichthys miiuy sperm cryopreservation were 10%Gly, 10%DMSO, 10%EG and 15%PG.
