声动力疗法对SD大鼠C6胶质瘤照射后单态氧含量的影响研究

摘要:目的:本文研究了声动力学疗法对大鼠胶质瘤照射后单态氧(1O2)含量的影响。材料与方法:取56只SD大鼠皮下注射C6胶质瘤细胞,待肿瘤直径达2cm,随机分成7组,每组8只,其中一组为对照组,于肿瘤内注射声敏剂血啉甲醚(HMME,10mg/kg),对照组未注射声敏剂。注射后给予声动力学疗法以功率为0.5W/cm2、频率为1MHz超声照射120s,将照射后大鼠分别于照射后即刻、6h, 12h, 24h, 48h, 72h 取出肿瘤样本,液氮冷冻储存,真空冷冻抽干,研磨后用电子自旋磁共振机测量单态氧含量。结果:在所选定的剂量下,照射后单态氧浓度逐渐增加,至24h时达到高峰,照射后48h以后单态氧含量显著减低。结论:声动力治疗作用对肿瘤的抑制途径之一是声动力治疗后单态氧的产生短时间内迅速升高,对肿瘤细胞起DNA声敏作用,作用于细胞膜,线粒体以及核酸,达到杀死肿瘤细胞目的。

Effect of sonodynamic therapy on singlet oxygen content of C6 glioma cells of SD rat after ultrasound radiation
Abstract:Objective:Content of C6 glioma cells of SD rat after ultrasound radiation was to be studied in this paper. Materials and Methods: Subcutaneous injection of C6 glioma cells was performed for 56 SD rats, after tumor reached 2 cm, these rats were randomly divided into 7 groups, 8 rats/group, among which one group was taken as control group, and sonosensitizer hematoporphyrin monomethyl ether (HMME, 10mg/kg) was injected into tumor with control group as exception. After injection, these rats were treated by sonotodynamic therapy with 120s ultrasound radiation of 0.5W/cm2 power and 1 MHz frequency, 0h, 6h, 12h, 24h, 48 and 72h after radiation, tumor samples were collected, frozen and storied in liquid nitrogen, and dried by vacuum freeze, their Singlet oxygen contents were determined by Electron Paramagnetic Resonance Spectrometer after being ground. Results: At selected dosage, single oxygen concentration increased gradually after irradiation and reached peak at 24 h and decreased, and single oxygen content decreased significantly after 48 h of irritation..Conclusion: One inhibitory path of sonotodynamic therapy for tumor is that yield of Singlet oxygen was improved rapidly in short time after sonotodynamic therapy, which had DNA soundsensitive effect on tumor, acted on cell membrane, mitochondria and nucleic acid and reached the aim of killing tumor cells.
