复频聚焦超声激活血卟啉单甲醚对体外鼠C6胶质瘤细胞的杀伤研究

摘要:目的:研究复频聚焦超声激活血卟啉单甲醚(HMME)对体外鼠C6胶质瘤细胞的杀伤作用,并与单频聚焦超声的作用效果进行比较性研究。方法:取处于对数生长期C6胶质瘤细胞系,细胞悬液浓度为4×104个/ml。将各组细胞悬浮液以每孔200μl (细胞浓度:4×104/ml)接种于洁净的96孔培养板上,培养板随机分为4组:(A) 对照组 (B)单频聚焦超声治疗组F1;(C) 单频聚焦超声治疗组F2;(D) 复频聚焦超声治疗组(F1+F2)。治疗组加入HMME终浓度为10μg/ml。各组避光2h,治疗组进行US照射.继续避光培养6、12、24。应用MTT法在上述时间点计算抑瘤率。结果:复频聚焦超声激活血卟啉单甲醚在各个时间点均能有效的杀伤C6胶质瘤细胞,与单频超声激活血卟啉单甲醚的作用效果相比,具有良好的协同作用。结论:我们的研究表明复频聚焦超声激活血卟啉单甲醚能够有效的增加体外C6胶质瘤细胞的杀伤率。

The killing effect of C6 glioma cells induced by hematoporphyrin monomethyl ether mediated sonodynamic treatments with bi-frequency focal ultrasound
Abstract:Objectives: To study the cytotoxicity of C6 glioma cells induced by hematoporphyrin monomethyl ethe(HMME) mediated sonodynamic treatments(SDT) with bi-frequency focal Ultrasound by compared with uni-frequency one. Methods: Equal number of exponential growth phase cells were transferred to 96-wells plates( Each well contained 200 μl cell suspension and approximately 4 × 104 cells/ml).The plates were divided randomly and evenly into 4 groups (control, uni-frequency ultrasound (F1), uni-frequency ultrasound (F2) and bi-frequency focal Ultrasound (F1+F2). Cells in treating groups were incubated with HMME (final concentration -10 μg/ml) in the dark for 2 h. The growth inhibition rate of C6 glioma cells at in different time points(6,12 and 24h)were investigated by MTT assay. Results: The growth inhibition rate of C6 glioma cells in bi-frequency focal ultrasound groups significantly increased at different time points. Bi-frequency focal Ultrasound and HMME had synergic effect on killing tumor cells compared with the uni-frequency one. Conclusions: Our preliminary data indicated that bi-frequency focal Ultrasound activing HMME could increasing killing rate of C6 glioma cells in vitro
