单因素考察及正交设计确定三氧化二砷PLGA纳米粒制备工艺

摘要:目的: 采用复乳法制备三氧化二砷纳米粒,研究不同水平的因素对三氧化二砷PLGA纳米粒的粒径、包封率的影响。方法:分别考察载体质量、有机相种类及体积、乳化剂种类及浓度、超声时间的各个水平、药物浓度、外水相浓度及体积,正交设计法及单因素考察确定最佳处方,并考察了交互影响。制备三氧化二砷PLGA纳米粒,分光光度法测定包封率,纳米粒度仪测定纳米粒的粒径。结果:以乙酸乙酯(2ml)为溶剂,2%的Poloxamer407为外水相,累计超声时间60s,得到平均粒径为83.2nm,包封率为88.67%的纳米粒。结论:PLGA以复乳法制备三氧化二砷纳米粒方法简单、方便、可行。

Single Factor Review and Orthogonal Design of Arsenic Trioxide Nanoparticles\\\\\\\
Abstract:Objective: To prepare arsenic trioxide nanoparticles with W/O/W(water/oil/water)double emulsion--solvent evaporation method, to study the effects on mean diameter and encapsulation with different levels of factors. Method: Choose the mass of carrier, the kinds and volume of organic phase, the kinds and consistence of emulsifier, the levels of ultrasound time, the drug consistence, the consistence and volume of outer water phase as review factors. Use orthogonal design and single factor review to confirm the optimum prescription and alternant affect. Use nanometer particles sizer and zata potential analyzer to determine the mean diameter, ultraviolet-visible spectrometer to determine the drug loading. Result: Use acetic ether(2ml)as solvent, 2% Poloxamer407 as outer water phase, and 60 seconds as accumulative ultrasound time. Get a mean diameter for the 83.2 nm, encapsulation rate is 88.67% of nanoparticles. Conclusion: The method of W/O/W is simple and convenient.
