水滴为模板制备蜂窝状表面的聚己内酯多孔膜及其细胞亲和性

摘要:种子细胞、支架材料、生长因子是构建组织工程化组织或器官的三要素。其中支架材料的表面物理形貌对细胞的黏附、增殖和分化等行为具有重要的影响。本文以水滴(Breath Figure)为模板制备了PCL蜂窝状表面多孔膜,用SEM研究了制备条件即环境相对湿度、温度和聚合物溶液浓度对孔大小和分布的影响。用细胞计数法和MTT法考察了兔的骨髓间充质干细胞(MSC)在多孔PCL膜表面的黏附效率和增殖行为。结果表明多孔膜表面孔的孔径随环境湿度增加而增加,随着溶液浓度增加而减小,随环境温度升高而减小,与平滑表面的PCL膜相比,蜂窝状的多孔表面有效地提高了细胞的黏附效率并促进了细胞的增殖。

Preparation of Honeycomb-Structured surfaces of polycaprolactone films and its cell affinity
Abstract:Seed cells, scaffold materials and growth factors are three elements to construct tissue engineered tissues and organs in tissue engineering. The surface topography of scaffold materials plays a crucial role in controlling cell behavior including cell adhesion, proliferation and differentiation. In this study, the honey-structured surfaces of PCL film were prepared by a molding technique using the water droplets as a template. The relationship between the pore size and environmental relative humidity, temperature and concentration of PCL was investigated by SEM observation. The cell adhesion, proliferation behavior and morphology on the honey-structured surfaces were characterized by SEM observation, cell attachment efficiency measurement and MTT assay, respectively. The results of SEM show that the pore diameter of the honey-structured films increases with the increase of relative humidity and decreases with the increase of temperature and concentration in a certain range. Cell culture shows that the cell adhesion and proliferation could be enhanced on PCL honey-structured surfaces compared to the smooth surfaces.
