实验性脾气虚对大鼠睾丸生精细胞凋亡的影响

摘要:目的:探讨大鼠脾气虚对睾丸生精细胞凋亡的影响,阐明脾气虚引起不育的病理机制。方法:选用雄性wistar大鼠40只,随机分为正常组、21d模型组、35d模型组及50d模型组。采用双因素复合法即苦寒泻下加劳倦过度法(大黄灌胃加游泳法)建立脾气虚模型,流式细胞术分别检测各组大鼠睾丸凋亡细胞数及各级生精细胞百分比。结果:(1)正常组出现低平的凋亡峰, 各模型组出现明显的凋亡峰, 且随着造模时间的延长凋亡峰增高。(2)各模型组1C和4C生精细胞百分比较正常组明显降低,以50d模型组为甚。结论:脾气虚可引起睾丸生精细胞大量凋亡,各级生精细胞数减少,这可能是脾气虚引起睾丸生精功能障碍从而导致不育的根本机制。

Effects of Experimental Spleen Qi deficiency on the Apoptosis of Spermatogenic Cells in Rats
Abstract:Objective:To explore the effects of experimental Spleen Qi deficiency on the apoptosis of spermatogenic cells in rats. Methods: 40 male wistar rats were randomly divided into normal group, 21d model group , 35d model group and model group 50d. 40 male wistar rats were randomly divided into normal group, 21d model, 35d model group and model group 50d. Method using two-factor complex that is tired and cold diarrhea excessive Act (administered rhubarb add swimming Law) set up spleen Qi Deficiency model, flow cytometry in each group were used to detect apoptotic cells in rat testis number and percentage of spermatogenic cells at all levels. Results: (1) Low marked apoptosis peak was observed in the control group, but it was ob-served in all the other groups, and the height of the peak increased with the duration of experiment.(2) the model group 1C and 4C spermatogenic cell percentage was significantly lower than normal, especially in the model group 50d. Conclusion: Spleen Qi Deficiency can induceapoptosis of large numbers of spermatogenic cells, and the decrease of spermatogenic cells. This may be the mechanism of Spleen Qi Deficiency causing male sterility.
