利用液相芯片技术研究DAZL基因单核苷酸多态性与男性无精症关系

摘要:目的 建立液相芯片(MuLti-Analyze Suspension Array, MASA)检测单核苷酸多态性(single nucleotide polymorphism SNP)的方法,并探讨SNP与男性无精症的相关性。方法 基于Luminex 技术平台,国内率先应用等位基因特异性延伸反应(allele specific primer extension, ASPE),并将此方法应用于DAZL(deleted in azoospermia like)基因SNP260A→G和SNP386A→G研究。结果 病例组和对照组均存在SNP260A→G和SNP386A→G多态性,两个突变统计学分析差异均无显著性(P>0.05)。结论 成功建立并应用该技术。因为具有高通量,高准确性,低成本,低时耗等优点,利用液相芯片检测SNP是该研究领域的一个重要突破,

Research on the Relationship Between the SNPs of DAZL and Male Azoospermia by MuLti-Analyze Suspension Array
Abstract:Objective To establish a new method to genotype SNP(single nucleotide polymorphism), and explore the relationship between SNP and male Azoospermia. Methods SNP260 and SNP386 of DAZL(deleted in azoospermia like)gene were genotyped by the assay allele specific primer extension (ASPE). ResuLts Both the SNP260A→G and SNP386A→G were found in the infertility and the control groups, and there were no significant difference found for both of the mutations (P>0.05). Conclusion Successfully established and applied this method. Because of its high-throughput, high accuracy and low cost, this method is a breakthrough for genotyping SNP .
