人肺腺癌细胞抗肿瘤药物渗透模型PAMPA的构建及参数的优化

摘要:背景与目的:PAMPA渗透模型在药学领域有广泛的应用。本研究旨在通过肿瘤细胞膜渗透模型的建立,对药物的细胞渗透性进行预测。方法:在胃肠道吸收的PAMPA模型基础上对磷脂比例、渗透时间等影响因素进行优化,发展一套具有抗肿瘤药物渗透性吸收的检测方法,从而构建肿瘤细胞膜渗透模型。将人工膜的表观渗透值与真实细胞的渗透值的相关性进行比对。结果:本实验用PAMPA模型测试16种抗肿瘤药物的渗透特性并且与药物对细胞的吸收值做了对比。结合正交试验,优化后渗透实验的相关度可达0.77。

Model establishment and parameter optimizing of antineoplastic parallel artificial membrane permeation assay in human lung adenocarcinoma cell line A549
Abstract:Backgroud and purpose: antineoplastic compounds absorption can depend simultaneously on three interrelated properties: permeability, solubility and pKa. The parallel artificial membrane permeability assay (PAMPA), as a passivepermeability screen, is a possible low-cost alternative to cellular models for the earliest absorption–distribution–metabolism–excretion primary screening of research compounds. Its popularity in the industry has risen rapidly. PAMPA is a useful tool in drug discovery for the prediction of oral absorption, brain penetration and for the development of structure-permeability relationships. This study was to examine state-of-the-art PAMPA methods and establish a tumor cell membrane model, and predict the cell antineoplastic absorption. Methods: Based on analyzing the standard gastrointestinal permeability model, the method of predicting antineoplastic permeability was advanced by experimental condition optimization, and the possible permeability models of tumor cell membrane was given. Results: Experimental PAMPA data generated for 16 commercial drugs was compared with cell absorption value, the result showed that the optimized correlation degree was 0.77. Conclusions: The parallel artificial membrane permeation assay (PAMPA) permits the fast determination of artificial membrane permeation properties of drugs, which will help to develop new anticarcinogen with better bioavailibility
