3,5-二硝基水杨酸法测定玉米秸秆还原糖含量

摘要:本文探讨DNS试剂添加量、显色时间、测定波长以及显色后放置时间对测量结果的影响。结果表明,DNS试剂的用量2.5ml,沸水浴显色时间5min,显色定容20min后,在482nm波长条件下测定,数据的准确性和稳定性较好。玉米秸秆最佳水解条件为功率300w、时间40min、硫酸浓度2%、固液比1:12.5、压强0.4Mpa,在此条件下水解后得到的还原糖占总的玉米秸秆20.67%

The Study on Determination of Reducing Sugar Content in the Crop Straw by Using 3,5-Dinitrosalicylic Acid Method
Abstract:The paper researches the effects on determination results by the factors,such as amount of DNS reagent developing time,wavelength and storage time were discussed. The results showed that on the conditions of reagent amount around 2.5ml, boiling bath time not less than 5 minutes, and standing for 20 minutes after developing, at the wave length of 482nm, the accurate results can be acquired. The optimal conditions by acid pretreatment were power were 300 watt, processing time 40 min, concentration of H2SO4 2%, the ratio of solid and liquid were 1:12.5 and the pressure 0.4Mpa, under this condition, the reducing sugar was 20.67%.
