青蒿素治疗狼疮性肾炎小鼠的作用机制

摘要:目的:探讨青蒿素对狼疮性肾炎小鼠的治疗作用及其作用机制。方法:通过诱导方法建立狼疮性肾炎小鼠模型,观察青蒿素组小鼠症状、24h尿蛋白定量变化,同时分别采用放射免疫法、EMSA、Western blotting、Northern印迹杂交及RT-PCR等检测技术测定小鼠血清TNF-α、IL-6水平及肾组织NF-kBp65蛋白、NF-kB活性、TGF-β1 mRNA表达情况。结果:与病理组比较,青蒿素组狼疮性肾炎小鼠症状明显改善、24小时尿蛋白定量明显降低,肾脏的病理损害得到显著减轻(P<0.01);与病理组比较,青蒿素可明显降低小鼠血清IL-6、TNF-α水平及抑制肾组织NF-kB活性、NF-kBp65蛋白、TGF-β1mRNA表达(P<0.05或P<0.01)。结论:青蒿素可明显降低狼疮性肾炎小鼠的24h尿蛋白定量,改善症状,其作用机制可能与降低血清IL-6、TNF-α水平,抑制肾组织NF-kB活性、NF-kBp65蛋白和TGF-β1mRNA表达有关。

Study the therapeutic mechanism of artemisinin on lupus nephritis mice
Abstract:Objective: To investigave the effects and mechanism of artemisinin on the lupus nephritis mice.Methods: Establishing the mice model of chronic graft-versus-host disease(cGVHD) through the method of induction, we observed the change of symptom and urine protein/24h, at the same time ,the levels of TNF-α、IL-6 in serum and the expression of NF-kBp65 protein 、NF-kB、TGF-β1 mRNA in renal tissues were detected using radio-immunityassay、electrophoretic mobility shift assay (EMSA), Western blot assay、Northern Blotting and the technique of RT-PCR individly.Results: Compared with model group , the improvement of symptom ,decrease of urine protein/24h and alleviate of the renal pathological lesion in lupus nephritis mice were obviously in Artemisinin group(P<0.01);compared with model group , Artemisinin could decrease the levels of TNF-α、IL-6 in serum and inhibit the expression of NF-kBp65 protein 、NF-kB、TGF-β1 mRNA in renal tissues(P<0.05 or P<0.01).Conclusion: Artemisinin is able to improve the symptom ,decrease urine protein/24h in lupus nephritis mice, The mechanism may have relation with decreasing the levels of TNF-α、IL-6 in serum and inhibiting the expression of NF-kBp65 protein 、NF-kB、TGF-β1 mRNA in renal tissues.
