黄曲霉毒素致犬肝损伤代谢组的GC-MS分析

摘要:运用代谢组学方法研究黄曲霉毒素(AF)对犬肝脏的损伤作用。通过气相色谱-质谱技术(GC-MS)分析AF中毒犬血浆中的代谢物谱,并运用主成分分析(PCA)方法对血浆内源性代谢物随中毒时间的变化情况进行识别。研究发现,随着AF中毒时间的延长,犬血浆的代谢物谱存在着一定的变化规律。血浆中的部分氨基酸含量有所上升,葡萄糖、胆固醇以及部分游离脂肪酸含量下降。这些代谢物的变化都与肝脏的损伤程度密切相关,且变化趋势与血液生化指标检测结果一致。研究结果表明,基于GC-MS的代谢组学技术结合PCA的方法能较全面地反应生物体的代谢状态,有助于AF中毒的早期诊断。

Metabonomic Study of Aflatoxin-Induced Liver injury in canine by GC-MS
Abstract:A metabonomic study for the aflatoxin induced liver injure in canine was described. Gas chromatography –mass spectrometry(GC-MS) technique was used to analyze the canine metabonome in plasma and principal component analysis was introduced for the pattern recognition of canine metabonome with the change of poisoning time. The result presented clear changing tendencies of metabolite profiles with the extention of aflatoxicosis time. The increased concentrations of several amino acids, as well as the decreased concentrations of glucose、cholesterol and some free fatty acids in plasma, could be closely related to the degree of liver damage. Changing tendencies of the metabolite profiles were conformed to the results of blood biochemical indictor. The results suggested that GC-MS based on metabonomic combined with the PCA can be more comprehensive response the metabolic state and contributed to early diagnosis of aflatoxicosis.
