多色荧光稀土纳米颗粒的制备与应用

摘要:由于稀土荧光化合物中只有Eu(III)与Tb(III)的某些化合物具有强的荧光发光性能,其荧光发光的颜色分别为红色(铕化合物,约615 nm)和绿色(铽化合物,约545 nm),限制了这类化合物在多色荧光标记生化分析中的应用。通过将Eu(III)及Tb(III)与配体4’-苯基-2,2’:6’,2”-联三吡啶-6,6”-二甲胺四乙酸的荧光配合物同时共价键合到纳米硅胶颗粒中并调节Eu(III)/Tb(III)配合物摩尔比的方法,制备出了可发出红、橙、黄、绿系列颜色荧光的纳米稀土荧光颗粒。将纳米荧光颗粒标记到链霉亲和素后用于人前列腺特异抗原及病原微生物蓝氏贾地鞭毛虫的时间分辨荧光测定,结果表明新型纳米稀土荧光颗粒可作为多色荧光标记物用于时间分辨荧光生化测定。

Preparation and application of multicolor luminescent lanthanide nanoparticles
Abstract:Because strongly fluorescent lanthanide compounds are mainly limited to Eu3+ and Tb3+ compounds with red (Eu, ~615 nm) and green (Tb, ~545 nm) emissions, the development and application of fluorescence bioassay technique using lanthanide-based multicolor fluorescent biolabels are rarely investigated. A series of lanthanide complexes-bound silica nanoparticles with red, orange, yellow and green fluorescence colors was prepared by co-binding different ratios of Eu3+/Tb3+ fluorescent complexes with the ligand (4’-phenyl-2,2’:6’,2”-terpyridine-6,6”-diyl) bis(methylenenitrilo) tetrakis (acetic acid) inside the silica nanoparticles. The nanoparticle-labeled streptavidin was prepared and used for time-resolved fluorescence detections of human prostate-specific antigen and an environmental pathogen Giardia lamblia. The results indicate that the new nanoparticles are useful to multicolor biolabels for time-resolved fluorescence bioassays.
