温州蜜柑与有籽甜橙体细胞杂种再生及分子鉴定

摘要:国庆1号温州蜜柑(Citrus unshiu Marc. cv. Guoqing No. 1)愈伤组织原生质体分别与红暗柳橙(C. sinensis Osbeck cv. ‘Red Anliu’)和桃叶橙(C. sinensis Osbeck cv. ‘Taoye’)叶肉原生质体电融合。经培养分别得到8棵和20棵再生植株,随机选取其中部分植株进行倍性鉴定表明:除国庆1号与红暗柳橙组合中有1棵为二倍体外,其它均为四倍体。SSR、cpSSR和CAPS分子标记分别鉴定细胞核、叶绿体及线粒体遗传物质的组成,表明再生四倍体均带有双亲核SSR的特征带,细胞质基因组均来自愈伤组织亲本,为异源体细胞杂种;而二倍体植株的细胞核和细胞质遗传物质均与愈伤组织亲本一致,为愈伤亲本再生植株。探讨了这些融合再生植株的潜在育种价值。

Production and molecular characterization of interspecific somatic hybrids between Satsuma mandarin and two seedy sweet oranges
Abstract:Citrus symmetric fusions between suspension protoplasts of Citrus unshiu Marc. cv. Guoqing No. 1 (G1) and mesophyll protoplasts isolated from ‘Red Anliu’ sweet orange (C. sinensis Osbeck cv. ‘Red Anliu’) or from ‘Taoye’ orange (C. sinensis Osbeck cv. ‘Taoye’) were conducted. Eight plants from G1 + ‘Red Anliu’ and 20 plants from G1 + ‘Taoye’ were regenerated respectively. Flow cytometry analysis of randomly selected four plants recovered from G1 + ‘Red Anliu’ sweet orange and five plants regenerated from G1 + ‘Taoye’ sweet orange showed that three plants of G1 + ‘Red Anliu’ and all five plants from G1 + ‘Taoye’ plants were tetraploid and the remaining one was diploid. Genetic compositions of regenerated plants were determined by nuclear SSR, chloroplast-SSR and mitochondrial CAPS analysis. It was confirmed that all tetraploid plants were allotetraploid somatic hybrids, with cpDNA and mtDNA from embryogenic callus parent and the diploid was a regenerated plant from G1. The potential of these novel allotetraploid somatic hybrids in citrus breeding was discussed.
