摘要:细胞凋亡,即程序性细胞死亡,具有高度调控性和关键性的进程,存在于所有的多细胞生物体中。它不仅涉及调控机制的细胞,而且还和一些疾病因素有关,如炎症、恶性肿瘤、自身免疫以及神经退行性病变。各种各样的毒素可以诱导细胞凋亡。具有致癌性的过渡金属,如铅、镉都能促进细胞凋亡与DNA碱基的改变、链断裂和基因重组。免疫抑制机制有助于铅诱导细胞凋亡,而镉,金属硫蛋白表达决定细胞是凋亡或坏死。因而,细胞凋亡紊乱和细胞积聚可能在一些严重金属诱导性疾病中起关键作用,包括肝毒性、肾毒性,神经毒性,自身免疫和致癌作用。了解铅镉引起的细胞凋亡对研究铅镉中毒的分子防治具有重要意义。

Recent developments on apoptosis of Lead and Cadmium
Abstract:Apoptosis, also known as programmed cell death is a highly regulated and crucial process found in all multicellular organisms. It is not only implicated in regulatory mechanisms of cells, but has been attributed to a number of diseases, i.e. inflammation, malignancy, autoimmunity and neurodegeneration. A variety of toxins can induce apoptosis. Carcinogenic transition metals, viz. cadmium, chromium and nickel promote apoptosis along with DNA base modifications, strand breaks and rearrangements. Immuno suppressive mechanisms contribute in lead-induced apoptosis. In the case of cadmium, metallothionein expression determines the choice between apoptosis and necrosis. Apoptosis may play a critical role in some of the most debilitating metal-induced afflictions including hepatotoxicity, renal toxicity, neurotoxicity, autoimmunity and carcinogenesis. An understanding of metal-induced apoptosis will be helpful in the development of preventive molecular strategies.
