利用表面层层自组装技术制备蛋白质分子印迹聚合物

摘要:本工作将层层自组装与分子印迹技术相结合,首先利用表面层层自组装技术,将牛血清白蛋白和壳聚糖固定在交联壳聚糖基材表面,再经环氧氯丙烷处理后,洗脱模板蛋白质,制备出牛血清白蛋白分子印迹聚合物。通过水接触角测试、环境扫描电镜及蛋白质吸附测试等对蛋白质印迹聚合物进行了分析。测试结果表明,与非印迹聚合物相比,蛋白质印迹聚合物表面变粗糙,水接触角明显下降,并且对模板蛋白质的吸附量提高了27.7倍。

The preparation of the protein imprinted chitosan by LBL self-assembly technique
Abstract:In this work, a novel molecularly imprinted polymer was prepared by LBL self-assembly technique. The negatively charged bovine serum albumin and positively charged chitosan were alternately deposited onto the crosslinked chitosan substrate. After crosslinked by epoxy chloropropane and eluted by SDS-HAc solution, the protein imprinted polymer was formed. The property of imprinted polymer was characterized by water contact angle, environmental scanning electron microscope, and protein adsorption experiment. The results showed that the surface of imprinted polymer became rough, the water contact angle of the surface evidently decreased, and the BSA adsorption increased 27.7 times compared with the non-imprinted polymer.
