彝药“麻补”抗肿瘤活性物质基础及作用机理研究

摘要:目的:对彝药“麻补”(狭叶重楼P.polyphylla.Smith var stenophylla Franch)抗肿瘤活性的物质基础及作用机理进行初步研究。方法:采用植物化学技术从“麻补”药材中分离鉴定出1种C27甾体皂苷类化合物--重楼皂苷Ⅵ(PSⅥ),采用MTT法测定了PSⅥ对人卵巢癌细胞株SKOV-3的增殖抑制活性以及对人卵巢上皮细胞、脑膜细胞、血管平滑肌细胞、支气管上皮细胞等正常组织细胞的细胞毒性;采用荧光染色技术检测了药物干预对肿瘤细胞毒性的作用机理;利用透射电镜观察给药后细胞的显微结构变化。结果:PSⅥ对上述4类正常组织细胞无显著细胞毒性;对人卵巢癌细胞株SKOV-3有细胞毒性,IC50为42.61μmol/L,其细胞毒性机理为诱导肿瘤细胞凋亡。

Research on the material basis and mechanism for anti-tumor activity of Yi medicine ”MaBu”
Abstract:Objective: To carry out elementary pharmacological research on the antitumor activity and mechanism of Yi medicine ”MaBu” (Paris. polyphylla Smith var. stenophylla f. latifolia(Wang et Tang)H. Li)from Liangshan Yi Autonomous Prefecture. Methods: One kind of C27 steroidal saponin from ”MaBu” was isolated and identified as ParissaponinⅥ(PSⅥ).The effect of proliferation inhibitory activity of PSⅥ on SKOV-3 ovarian tumor cell and cytotoxicity of PSⅥ on human ovarian surface epithelial cells , human meningeal cells, vascular smooth muscle cells and bronchial epithelial cells were measured by MTT method. The mechanism of cytotoxicity effect of PSⅥ on tumor cell was disclosed by Fluorescent staining method. The microstructure change inside SKOV-3 ovarian tumor cell with treatment of PSⅥ was observed by Transmission Electron Microscopic(TEM). Results: PSⅥ shows insignificant cytotoxicity effect on 4 kinds of normal tissue cell mentioned above and , however, significant cytotoxicity effect on SKOV-3 ovarian tumor cell with IC50 42.61μmol/L. Induced apoptosis is involved in the mechanism of cytotoxicity effect of PSⅥ on tumor cell.
